TOS propionate agar medium is specifically prepared for selective enumeration of Bifidobacteria in fermented milks and fermented milk drinks living together with Lactic acid bacteria.

TOS propionate agar medium contains highly purified "Galactooligosaccharides", which is one of the most excellent Bifidobacteria growth substances.

Its pH adjustment is not necessary and for sterilization autoclave at 115Ž for 15minutes.
Anaerobic chamber, Anaerobic jar (Steal wool method) and commercial simple Anaerobic kit are able to use for Anaerobic culture.

Dissolve 62.5g in final 1,000ml of distilled water and autoclave at 115Ž for 15min.
‡A Place approximately 15-ml aliquots in a sterile Petri dish(90mm„U)and cool down for gelling.
‡B Spread 0.1ml of serially diluted cell suspensions over the surface of the plate with a spreader.
™Caution Avoid inhalation during weighing powdered medium. Do not use for other purposes.

Since the colonies of lactic acid bacteria are generally smaller in size
(pinpoint colonies) than those of Bifidobacteria, you can easily distinguish
the colonies between them.
